بررسی اثرات مجزا و ترکیبی فلوکستین و ریزپلاستیک‌های پلی‌استایرن بر پارامترهای خون‌شناسی ماهی قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss)

چکیده
محیط‌های آبی به‌طور فزاینده‌ای تحت تأثیر آلاینده‌های نوظهور نظیر پسماندهای پلاستیکی و دارویی قرار دارند. فلوکستین (FLX)، یک داروی ضدافسردگی پرمصرف، و ریزپلاستیک‌های پلی‌استایرن (PS)، از فراوان‌ترین آلاینده‌های پایدار، به‌دلیل پایداری بالا و اثرات بالقوه مخرب، تهدیدی جدی برای سلامت آبزیان محسوب می‌شوند. با توجه به اینکه ارگانیسم‌ها در محیط طبیعی به‌ندرت در معرض یک آلاینده‌ منفرد قرار می‌گیرند، این مطالعه با هدف ارزیابی اثرات سم‌شناختی مجزا و ترکیبی نانوپلاستیک و میکروپلاستیک پلی‌استایرن و داروی فلوکستین بر شاخص‌های خون‌شناختی بچه‌ماهی قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss) طراحی گردید. بچه‌ماهیان قزل‌آلای رنگین‌کمان به‌مدت ۲۸ روز با جیره‌های غذایی حاوی فلوکستین (۷/۶ میلی‌گرم بر کیلوگرم)، نانوپلاستیک پلی‌استایرن (۱۰۰ نانومتر؛ در دو غلظت ۵۰۰ و ۱۰۰۰ میلی‌گرم بر کیلوگرم)، میکروپلاستیک پلی‌استایرن (7 میکرومتر؛ در دو غلظت ۵۰۰ و ۱۰۰۰ میلی‌گرم بر کیلوگرم) و ترکیبات آنها تغذیه شدند. در پایان دوره، نمونه‌های خون جمع‌آوری و پارامترهایی شامل تعداد گلبول‌های قرمز (RBC)، تعداد گلبول‌های سفید (WBC)، درصد هماتوکریت (Hct)، غلظت هموگلوبین (Hb) و درصد افتراقی لکوسیت‌ها اندازه‌گیری شدند. نتایج نشان داد که مواجهه با آلاینده‌ها، به‌ویژه در تیمارهای ترکیبی، اثرات نامطلوب معنی‌داری بر پروفایل خونی ماهیان داشت. تعداد گلبول‌های قرمز، هموگلوبین و هماتوکریت در بیشتر گروه‌های تیماری نسبت به گروه شاهد کاهش معنی‌داری یافتند (0/05>P). تعداد گلبول‌های سفید در گروه فلوکستین و گروه‌های ترکیبی افزایش معنی‌داری نشان داد، اما در گروه‌های پلاستیک تغییر معنی‌داری مشاهده نشد (0/05<P). همچنین، تغییرات معنی‌داری در درصد افتراقی لکوسیت‌ها، شامل افزایش نوتروفیل‌ها و کاهش لنفوسیت‌ها، به‌ویژه در بالاترین دوز مواجهه ترکیبی، ثبت گردید (0/05>P). این مطالعه نشان داد که فلوکستین و پلاستیک‌های پلی‌استایرن، هم به‌صورت مجزا و هم ترکیبی، سلامت خون‌شناختی ماهی را مختل می‌کنند. تغییرات مشاهده‌شده بیانگر بروز استرس فیزیولوژیک سیستمیک، اختلال در ظرفیت حمل اکسیژن و تعدیل پاسخ‌های ایمنی است. این یافته‌ها بر ضرورت ارزیابی جامع خطرات زیست‌محیطی با در نظر گرفتن تعاملات پیچیده میان آلاینده‌های نوظهور در اکوسیستم‌های آبی تأکید دارد.

Aquatic environments are increasingly impacted by emerging contaminants such as pharmaceutical residues and plastic waste. Fluoxetine (FLX), a widely prescribed antidepressant, and polystyrene (PS-MNPs), among the most abundant persistent pollutants, pose significant threats to aquatic health due to their high stability and potential detrimental effects. Given that organisms in natural environments are rarely exposed to a single contaminant, this study was designed to evaluate the individual and combined toxicological effects of polystyrene nanoplastics (PS-NPs), microplastics (PS-MPs), and fluoxetine on hematological indices in juvenile rainbow trout (Oncorhynchus mykiss). For 28 days, juvenile trout were fed diets containing fluoxetine (6.7 mg/kg), PS-NPs (100 nm; at 500 and 1000 mg/kg), PS-MPs (7 μm; at 500 and 1000 mg/kg), and their combinations. Following the exposure period, blood samples were collected, and parameters including red blood cell (RBC), white blood cell (WBC), hematocrit (Hct), hemoglobin (Hb), and differential leukocyte count were measured. Results indicated that exposure to the contaminants, particularly in combined treatments, induced significant adverse effects on the hematological profile. RBC, Hb, and Hct exhibited significant decreases in most treatment groups compared to the control (P<0.05). WBC increased significantly in the FLX and combined treatment groups, but no significant change was observed in the plastic-only groups (P>0.05). Significant alterations in differential leukocyte counts were also recorded, including increased neutrophils and decreased lymphocytes, especially at the highest combined exposure dose (P<0.05). This study demonstrates that FLX and PS-MNPs, both individually and in combination, disrupt the hematological health of fish. The observed changes indicate systemic physiological stress, impaired oxygen-carrying capacity, and immunomodulatory effects. These findings underscore the necessity for comprehensive ecological risk assessments that account for the complex interactions among emerging contaminants in aquatic ecosystems.
