تأثیرعصاره الکلی بره موم زنبورعسل پرتوتابی شده با اشعه گاما بر فاکتورهای رشد و ایمنی میگوی پاسفید غربی (Litopenaeus vannamei)

چکیده
این مطالعه به بررسی تأثیر عصاره الکلی بره موم زنبورعسل پرتوتابی شده با اشعه گاما بر برخی پارامترهای رشد و همولنف میگوی پا سفید غربی (Litopenaeus vannamei) پرداخت. آزمایش به‌صورت کاملاً تصادفی و در قالب 7 تیمار با 3 تکرار طی یک دوره 42 روزه انجام شد. تعداد 1050 پست لارو میگو با وزن اولیه 1-0/5 گرم تیماربندی شده و با سطوح مختلف عصاره الکلی بره موم در غلظت‌های صفر (شاهد)، 0/5 و 1 درصد در هر کیلوگرم جیره غذایی، پرتوتابی شده با دوزهای 5، 10 و 30 کیلوگری اشعه گاما تغذیه شدند. در آغاز و پایان دوره، از هر تکرار 5 قطعه میگو به‌طور تصادفی صید و پارامترهای مختلف از جمله وزن متوسط، افزایش وزن بدن (GR)، درصد افزایش وزن (WG)، ضریب رشد ویژه (SGR) و ضریب تبدیل غذایی (FCR) محاسبه شد. همولنف نیز به‌منظور بررسی تعداد و نوع هموسیت‌ها از حفره پریکاردیال جمع‌آوری گردید. نتایج نشان داد که استفاده از عصاره بره موم پرتوتابی شده با اشعه گاما به‌طور معنی‌دار موجب افزایش نرخ بازماندگی میگوی وانامی در شرایط آزمایشگاهی شد (0/05>P). شاخص‌های وزن نهایی و به‌تبع آن GR و BWI در تیمارهای 3 (غلظت 0/5 % با دوز 10 کیلوگری) و 4 (غلظت 1% با دوز 10 کیلوگری) و همچنین تیمار 6 (غلظت 1% با دوز 30 کیلوگری) به‌طور معنی‌دار نسبت به گروه شاهد افزایش یافت (0/05>P). شاخصFCR  تنها در تیمار 4 به‌طور معنی‌داری کمتر از گروه شاهد بود. بررسی همولنف نشان داد که بین میزان هموسیت کل در گروه شاهد و تیمارهای آزمایش اختلاف معنی‌داری وجود نداشت (0/05<P). با این حال، بیشترین مقدار سلول‌های دانه‌دار بزرگ و سلول‌های نیمه‌دار در تیمار ۴ مشاهده شد که از نظر آماری با گروه شاهد تفاوت داشت (0/05>P). همچنین، بیشترین میزان سلول‌های هیالین در گروه شاهد مشاهده گردید که از نظر آماری با تیمارهای دیگر تفاوت معنی‌داری نداشت. با توجه به نتایج به‌دست‌آمده، می‌توان نتیجه‌گیری کرد که مناسب‌ترین غلظت عصاره الکلی بره موم در جیره برای تحریک فاکتورهای رشد و ایمنی میگوی پا سفید غربی، غلظت 1% پرتوتابی شده با دوز 10 کیلوگری اشعه گاما می‌باشد.

The effect of gamma-irradiated bee propolis alcoholic extract on growth and immune factors of Pacific white shrimp (Litopenaeus vannamei)

چکیده English

This study investigated the effect of alcoholic propolis extract, irradiated with gamma rays, on growth parameters and hemolymph of Pacific white shrimp (Litopenaeus vannamei). The experiment was conducted in a completely randomized design with 7 treatments and 3 replications over a period of 42 days. A total of 1,050 post-larval shrimp with an initial weight of 0.5-1 grams were assigned to treatments with different levels of alcoholic propolis extract at concentrations of 0% (control), 0.5%, and 1% per kilogram of feed, irradiated with doses of 5, 10, and 30 kilograys of gamma rays. At the beginning and end of the experiment, 5 shrimp were randomly sampled from each replication to measure various parameters, including average weight, growth rate (GR), percentage weight gain (WG), specific growth rate (SGR), and feed conversion ratio (FCR). Hemolymph was collected from the pericardial cavity to assess the number and type of hemocytes. The results indicated that the use of gamma-irradiated propolis extract significantly increased the survival rate of vannamei shrimp under laboratory conditions (P<0.05). Final weight indices, and consequently GR and body weight increase (BWI), were significantly higher in treatments 3 (0.5% concentration with 10 kilogray dose), 4 (1% concentration with 10 kilogray dose), and treatment 6 (1% concentration with 30 kilogray dose) compared to the control group (P<0.05). FCR was significantly lower only in treatment 4 compared to the control group. Hemolymph analysis showed no significant difference in total hemocyte count between the control and experimental treatments (P>0.05). However, the highest levels of large granular cells and semi-granular cells were observed in treatment 4, showing a significant difference from the control group (P<0.05). The highest amount of hyaline cells was found in the control group, with no significant difference from other treatments. Based on the obtained results, it can be concluded that the most suitable concentration of alcoholic propolis extract in the diet for stimulating growth and immune factors in Pacific white shrimp is 1% irradiated with a 10 kilogray gamma dose.
