بررسی عملکرد رشد و آنزیم‌های گوارشی در میگوی پاسفید غربی (Litopenaeusvannmei) تغذیه‌شده با عصاره ترکیبی ماکروجلبک‌های قهوه‌ای و پروبیوتیک تک‌سل (Bacillus subtilis IS0)

چکیده
مطالعه اثرات عصاره ماکروجلبک و پروبیوتیک‌های گونه باسیلوس در میگوی پاسفیدغربی برای بهبود شیوه‌های آبزی‌پروری، بهبود سلامت و رشد میگو، مدیریت مؤثر بیماری‌ها و ارتقای پایداری در صنعت حیاتی است. بنابراین در این تحقیق به بررسی اثر توأم عصاره ترکیبی ماکروجلبک‌های قهوه‌ای (Padina astraulis،  Sargassum ilicifolium و Stoechospermum marginatum) و پروبیوتیک تک‌سل (Bacillussubtilis IS0) بر عملکرد رشد و فعالیت‌‌ آنزیم‌های گوارشی میگوی پاسفید غربی پرداخته شده است. در این مطالعه، 2400 پست لاروها با میانگین وزنی 1/02±28/9میلی‌گرم به‌طور تصادفی با تراکم 100 قطعه در هر واحد آزمایش توزیع شدند. هشت تیمار شامل: گروه شاهد از جیره غذایی فاقد عصاره ترکیبی ماکروجلبک (MPE) و پروبیوتیک تک‌سل (P) و بقیه گروه‌ها به‌ترتیب از جیره غذایی حاوی 15 گرم بر کیلوگرم غذا عصاره ترکیبی ماکروجلبک‌ها (MPE)، 1 گرم 100 گرم غذا پروبیوتیک تک‌سل (1P)، 2 گرم 100 گرم غذا پروبیوتیک تک‌سل (2P)، 3 گرم 100 گرم غذا پروبیوتیک تک‌سل (3P)، عصاره ترکیبی ماکروجلبک‌ها با غلظت 1گرم بر 100 گرم غذا پروبیوتیک تک‌سل (1P+MPE)، عصاره ترکیبی ماکروجلبک‌ها با غلظت 2 گرم بر 100 گرم غذا پروبیوتیک تک‌سل (2P+MPE) و تیمار حاوی عصاره ترکیبی ماکروجلبک‌ها با غلظت 3 گرم بر 100 گرم غذا پروبیوتیک (3P+MPE) به‌مدت60 روز تغذیه شدند. از کیت‌ها و دستگاه اسپکتروفتومتر برای سنجش فعالیت آنزیم‌های گوارشی استفاده شد. نتایج نشان داد که بیشترین میزان ضریب رشد ویژه، وزن نهایی، وزن به‌دست آمده در گروه‌های تغذیه‌شده با 2P+MPE و 3P+MPE مشاهده شد که اختلاف معنی‌داری را با گروه شاهد و بقیه گروه‌های مورد آزمایش نشان داد (0/05>P). کمترین میزان ضریب تبدیل غذایی در گروه‌‌های تغذیه‌شده با 2P+MPE و 3P+MPE مشاهده شد. میزان بقاء در بین گروه‌های مورد آزمایش تفاوت معنی‌داری را بایکدیگر نشان نداد (0/05<P). بیشترین میزان فعالیت آنزیم‌های لیپاز (0/11±8/33 واحد بر میلی‌گرم پروتئین)، آمیلاز (15/27±543/33 واحد بر میلی‌گرم پروتئین) و پروتئاز (10/21±488/66 واحد بر میلی‌گرم پروتئین) در گروه تغذیه‌شده با 2P+MPE مشاهده شد (0/05>P). به‌طورکلی می‌توان نتیجه گرفت که استفاده توأم عصاره ترکیبی ماکروجلبک‌های قهوه‌ای (P. astraulis، S. ilicifolium و S.marginatum) با غلظت‌های 2 گرم بر 100 گرم غذا، باعث بهبود عملکرد رشد و فعالیت آنزیم‌های گوارشی در میگوی پا سفید می‌شود.

The study of the effects of macroalgae extract and probiotics of the Bacillus species on the Pacific white shrimp for improving aquaculture practices, enhancing shrimp health and growth, effectively managing diseases, and promoting sustainability in the industry is vital. These findings could lead to significant advancements in shrimp farming and contribute to the overall success of aquaculture. This research examines the combined effect of a blend of brown macroalgae extracts (Padina astraulis, Sargassum ilicifolium, and Stoechospermum marginatum) and the single-cell probiotic (Bacillus subtilis IS0) on the growth performance and digestive enzyme activity of whiteleg shrimp, Litopenaus vannamei. A total of 2400 post-larvae with an average weight of 1.02±28.9 mg were randomly divided into eight groups of 100 shrimp each (with three replicates per group). The control group received a diet without macroalgae extract (MPE) or probiotic (P), while the other groups received diets containing different amounts of macroalgae extract (15 kg-1) and different concentrations of probiotic (1, 2 and 3 g 100 g-1 diet). Some groups also received combinations of the macroalgae extract and the probiotic at fixed concentrations. This feeding regimen lasted for 60 days. Spectrophotometer kits and devices were used to measure the activity of digestive enzymes.The results showed that the groups fed with P2+MPE and P3+MPE had the highest specific growth rates, final weights and weight gains, showing significant differences when compared with the control group and other experimental groups (P<0.05).In addition, these two groups also had the lowest feed conversion ratios. However, the survival rates of the experimental groups were not significantly different (P>0.05). The highest enzyme activities were found in the group receiving P2+MPE, with lipase at 8.33±11.0 U mg-1 protein, amylase at 543.33±15.27 U mg-1 protein and protease at 488.66±10.21 U mg-1 protein (P<0.05). Overall, the combined use of brown macroalgae extract at a concentration of 2 g 100 g-1 diet resulted in improved growth performance and increased digestive enzyme activity in white leg shrimp.
