بررسی اثرات سطوح مختلف نانوپلاستیک پلی‌استایرن بر بیان برخی ژن‌های مرتبط با سیستم ایمنی ماهی قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss)

چکیده

بسیاری از مواد پلاستیکی که در حال حاضر برای بسته‌بندی مواد غذایی استفاده می‌شوند، غیر قابل تجزیه بوده یا فرآیند تجزیة بسیار طولانی دارند، که باعث افزایش آلودگی محیط زیست می‌شوند. این تحقیق، با هدف بررسی اثرات سمی نانوپلاستیک پلی‌استایرن NPsPS به‌صورت انتقال از جیره غذایی به بدن بچه ­ماهیان قزل­ آلای رنگین‌کمان (Oncorhynchus mykiss) انجام گردید. برای این تحقیق، چهار تیمار از غلظت­ های NPsPS در غذا طبق تحقیقات انجام شده با مقادیر 0 (شاهد)، 0/1، 10 و 100 در نظر گرفته و در یک دوره زمانی مزمن 28 روزه، پس از گذشت یک هفته دوره سازگاری، در سه تکرار (در هر تکرار از هر غلظت، 15 قطعه بچه ­ماهی)، طراحی و اجرا گردید. بررسی تغییرات دو زیست‌نشانگر ژنی شاخص سیستم ایمنی و استرس در پروفایل بیانی بافت کبد و طحال، نشان داد که به‌دنبال افزایش غلظت NPsPS، سطوح بیان ژن­ های پروتئین شوک حرارتی 70 (HSP70) و فاکتور نکروز توموری آلفا (TNF-a) در هر دو بافت کبد و طحال، با افزایش رونویسی mRNA افزایش معنی ­داری داشته است (0/05>P). براساس نتایج به‌دست آمده، رهایش NPsPS برای سلامت و ایمنی در سطح سلولی-مولکولی در نمونه آبزی مورد پژوهش، تأیید شد و احتمال می ­رود در صورت رهایش این نانو ماده در محیط زیست آبی، القاگر اثرات نامطلوب زیست‌محیطی باشد.

چکیده [English]

Most of the plastic materials used for food packaging are non-biodegradable, resulting in more environmental pollution. Hence, this study was designed and conducted to measure the effect of toxicity of the PSNPs while transferring from the diet to the body of the fish (food concentrations = 0, 0.1, 10, and 100 mg/kg) on juvenile rainbow trout (Oncorhynchus mykiss) in a 28-day chronic treatment and the control group (food concentration= 0mg/kg) after the 7-day adaptation period, in three repetitions (15 juvenile trout were included in each repetition per concentration). In addition, changes in two genetic biomarkers of the immune system and stress in the expression profile of the liver and spleen tissues indicated that following the increase in the concentration of PSNPs, the tumor necrosis factor-alpha (TNF-α) and expression level of heat shock protein 70 (HSP70) in both liver and spleen tissues significantly increased with the increase of mRNA transcription (P<0.05). Based on the obtained results, the release of NPsPS was confirmed for health and safety at the cellular-molecular level in the aquatic sample under study, and it is likely that if this nanomaterial is released in the aquatic environment, it will induce adverse environmental effects.
