مطالعه تغییرات شاخص‌های فیزیکی و بقا ‌تخمک قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss) نگهداری شده در محیط های مختلف طی چند دوره زمانی

چکیده

در این مطالعه نگهداری کوتاه‌مدت تخمک قزل‌آلای رنگین‌کمان (Oncorhynchus mykiss) در محیط‌ها و دوره ­های زمانی مختلف بررسی شد. برای این منظور تخمک ­ها برای مدت زمان یک، چهار و هفت روز در دمای یخچال (4 درجه سانتی‌گراد) و در محلول­ های مایع تخمدانی، کورتلند بافرشده با Hepes و کورتلند بافرشده Tris-HCl نگه­داری شد؛ هر تیمار محلول-دوره زمانی در سه تکرار 10 گرمی انجام شد. شاخص‌های مورد بررسی شامل وزن‌تر، وزن خشک، قطر تخمک، pH مایع نگه­داری، درصد لقاح، درصد چشم­زدگی و درصد تفریخ بود. نتایج نشان داد که طول دوره نگه­داری تخمک اثر معنی‌داری بر وزن تر و خشک نداشته است اما بر قطر تخمک اثر‌گذار است و باعث افزایش آن می‌شود (0/05>P). میزان pH با افزایش مدت زمان نگه­داری به‌طور معنی‌داری در همه محیط‌ها کاهش یافت (0/05>P). بررسی درصد بقا تا چشم­ زدگی و تفریخ نشان داد که بیشترین بقا مربوط به تخمک نگه­داری شده در محیط ‌Hepes بود. از این ­رو می­ توان محیط بافری تولید شده با Hepes را به‌عنوان بهترین محیط مصنوعی برای نگهداری تخمک در قزل ­آلای رنگین­ کمان در کوتاه ­مدت معرفی کرد.

چکیده [English]

In this study, short-term storage of rainbow trout (Oncorhynchus mykiss) eggs in different environments and time periods was investigated. For this purpose, fish eggs were stored for one, four and seven days at refrigerator temperature (4°C) and in the natural ovarian fluid, Cortland buffered with Hepes and Tris-HCl, at pH=8.4. The individual egg wet and dry weights, egg diameter, storage fluid pH, fertilization, eyed and hatching percentage were measured. The results showed that egg storage time did not change wet and dry weights significantly, while egg diameter were increased significantly (P<0.05). The pH level decreased significantly with increasing storage time in all environments (P<0.05) although the highest decrease were observed in ovarian fluid,  Survival rates including fertilization, eyed and  hatching rates  were measured and indicated that the highest survival rates were obtained for eggs stored in Hepes environment. So, the artificial medium buffered with Hepes could be recommended for short-term storage of rainbow trout eggs.
