This study aimed to identify halophyte microalgae living in the back-barrier Lipar located on the shores of the Oman Sea in Sistan and Baluchestan Province. Water and phytoplankton sampling was carried out from the back-barrier in 2019-2020. Isolation and purification of the collected microalgae were performed by serial dilution and F2 medium culture in agar and liquid media, and finally, pure single colonies were obtained. Then, the initial identification of isolates was done through microscopic observations and comparison with morphologically valid classification keys. In the next step, 18S rRNA and 16S rRNA genes were used for further verification and identification by molecular method and sequencing. Analysis of morphological data and molecular sequencing showed that two purified isolates were green microalgae Dunaliella salina and green-blue microalgae Cyanothece sp., which was identified for the first time from the back-barrier. In terms of biological and commercial capacity, these two microalgae are suitable species for mass production and are widely used in industry. The purified strains have been donated to the National Bank of Iran Genetic Resources for future research or industrial-scale exploitation.}, keywords = {Microalgae,Lipar back barrier,sequencing,morphology,Cyanothece sp,Dunaliella Salina}, title_fa = {جداسازی، خالص‌سازی و شناسایی ریختی و مولکولی دو گونه ریزجلبک نمک‌دوست از کشندان پشت سدی لیپار (سواحل دریای عمان، چابهار)}, abstract_fa = {جداسازی و شناسایی میکروارگانیسم ­های ناشناخته بومی کشورمان و حفظ و نگهداری از آن‌ها در کلکسیون­ های ذخایر ژنتیکی به‌منظور بهره‌برداری از ظرفیت ­های زیستی آن‌ها از اهمیت و ضرورت ویژه‌ای برخوردار است. این مطالعه با هدف شناخت ریزجلبک­‌های نمک‌دوست ساکن در کشندان پشت سدی لیپار واقع در سواحل دریای عمان در استان سیستان و بلوچستان انجام شد. نمونه‌برداری از آب و فیتوپلانکتون­ های کشندان پشت سدی در سال ­های 1399-1398 صورت گرفت. جداسازی و خالص‌سازی ریزجلبک­ های جمع آوری شده با روش‌ تهیه‌ رقت­‌های سریالی و کشت در محیط­ های کشت آگار و محیط کشت مایع F2 به صورت متوالی صورت گرفته و در نهایت تک کلونی‌­های خالص به دست آمد. شناسایی اولیه جدایه‌ها از طریق مشاهدات میکروسکوپی و مقایسه با کلید­های طبقه‌بندی معتبر از لحاظ ریخت‌شناسی انجام شد. برای تایید و صحت بیشتر از شناسایی با روش مولکولی و توالی یابی ژن 18S rRNA و 16S rRNA استفاده شد. آنالیز داده‌های ریخت‌شناسی و توالی‌یابی مولکولی نشان داد که دو جدایه خالص شده ریزجلبک سبز Dunaliella salina و ریزجلبک سبز-آبی .Cyanothece sp می‌باشند که برای نخستین بار از کشندان پشت سدی شناسایی شدند. این دو ریزجلبک از نظر ظرفیت زیستی و تجاری جزء گونه‌های مناسب برای تولید انبوه و بسیار پرکاربرد در صنعت شناخته شده‌اند. سویه‌های خالص شده جهت نگهداری به بانک ملی ذخایر ژنتیک ایران اهدا شده تا جهت استفاده در تحقیقات و یا بهره‌برداری در مقیاس صنعتی در آینده به کار گرفته شوند.}, keywords_fa = {ریزجلبک,کشندان پشت سدی لیپار,توالی‌یابی,ریخت‌شناسی,Cyanothece sp,Dunaliella Salina}, url = {http://www.aquaculturesciences.ir/article_149610.html}, eprint = {http://www.aquaculturesciences.ir/article_149610_3233242c6ea071d8a769a94e75697060.pdf} }
