The results of extraction efficiency showed that the enzymatic method was three times more effective than acidic method. FTIR and SEM tests were used to further investigate the obtained collagens. The results showed that amides A and B appeared in similar regions for the obtained samples. Also, enzymes extracted by enzymatic method (PSC) had a more regular structure than collagen extracted by acidic method (ASC). The electrophoresis pattern of the extracted collagens revealed that the obtained collagens were type I and consisted of two 1α chains and one 2α chain with a density of 1α greater than 2α. The results of the percentage of neutralization of DPPH free radicals of the obtained collagens showed that among the concentrations of 0.1, 0.01 and 0.001 the highest concentration had the best performance and for ASC sample 34.35% and in the sample (ASCH) It was 53.86 % (P<0.05). Examination of emulsifying properties (EAI) of the obtained samples showed that both samples in acidic conditions showed higher emulsifying activity, the highest value of this index in acidic collagen 67.48% and enzymatic collagen 32/17%, respectively (P<0.05). The results of emulsion stability showed that hydrolyzed collagen samples at all pHs showed higher performance than non-hydrolyzed samples (P<0.05). The solubility of the samples was different at different pHs and the highest solubility of the samples was acidic pH with 2.38% for acidic collagen and 5.3% for enzymatic collagen (P<0.05). The results showed that the enzymatic method can have a better performance in extracting collagen from the skin of Scarus ghobban fish and also the obtained collagen can be used for food and cosmetic applications.}, keywords = {Parrot,collagen,DPPH,Functional Properties}, title_fa = {ارزیابی تاثیر روش‌های استخراج اسیدی و آنزیمی در بازیابی کلاژن از پوست طوطی ماهی (Scarus ghobban) و بررسی ویژگی‌های ساختاری، شیمیایی، ضداکسیدانی و عملکردی آن‌ها}, abstract_fa = {در پژوهش حاضر تاثیر روش ­های استخراج اسیدی و آنزیمی کلاژن از پوست طوطی ماهی مورد بررسی قرار گرفت. نتایج بازده استخراج نشان داد که در روش آنزیمی مقدار بازده استخراج سه برابر روش اسیدی بود. به منظور بررسی بیشتر کلاژن‌های به‌دست آمده تست ­های FTIR و SEM مورد استفاده قرار گرفت که نتایج حاکی از آن بود که آمیدهای A و  Bدر نواحی مشابهی برای نمونه­های به‌دست آمده ظاهر شده بودند و همچنین کلاژن استخراج شده به روش آنزیمی (PSC) در مقایسه با کلاژن استخراج شده به روش اسیدی (ASC) ساختار منظم تری داشت. الگوی الکتروفورز کلاژن‌های استخراج شده مشخص کرد که کلاژن­ های به‌دست آمده نوع I بوده و شامل دو زنجیره 1α و یک زنجیره 2α که دانسیته 1α بیشتر از 2α می‌باشد. نتایج درصد خنثی کنندگی رادیکال­های آزاد DPPH کلاژن­ های به‌دست آمده نشان داد که در بین غلظت‌های 0/1-0/01-0/001بالاترین غلظت بهترین عملکرد را داشتند و برای نمونه (ASC) 34/35 % و در نمونه (ASCH) 53/86 % بود (0/05>P). بررسی خواص امولیسیون کنندگی نمونه­ های به‌دست آمده مشخص کرد که در شرایط اسیدی در هر دو نمونه فعالیت امولسیون‌کنندگی بالاتری مشاهده شد که بالاترین مقدار آن در کلاژن اسیدی 67/48% و در کلاژن آنزیمی 17/32% بود (0/05>P). نتایج پایداری امولیسیون نشان داد که نمونه ­های کلاژن هیدرولیز شده در تمامی pH ها عملکرد بالاتری را نسبت به نمونه ­های هیدرولیز نشده داشت‌ (0/05>P). حلالیت نمونه­ ها در pH های مختلف متفاوت بود و بالاترین حلالیت نمونه ­ها در pH اسیدی با مقدار 2/38% برای کلاژن اسیدی و 5/3% برای کلاژن آنزیمی بود (0/05>P). نتایج نشان داد که روش آنزیمی می ­تواند عملکرد مناسب­تری در استخراج کلاژن از پوست طوطی ماهی داشته باشد. }, keywords_fa = {طوطی ماهی,کلاژن,  DPPH,خواص کارکردی}, url = {http://www.aquaculturesciences.ir/article_137671.html}, eprint = {http://www.aquaculturesciences.ir/article_137671_cbf7636675cdf73be31dbc027e0ee253.pdf} }
